Abstract
Amanita javanica is an edible mushroom from the subgenus Amanita, section Caesareae, that is widespread across Asia but remains poorly documented in several regions. In Kendari, Southeast Sulawesi, Indonesia, a yellow mushroom locally known as "Tanggarebonata" is widely consumed and sold, exhibiting morphological similarity to A. javanica. This study aims to confirm its identity using a combination of morphological, microscopic, molecular, and GC-MS analysis. Macroscopically, the specimen was examined directly from fresh samples preserved in 70% ethanol, showing typical Amanita features, including a yellowish pileus, free and crowded lamellae, a bulbous stipe base with a saccate volva, and the absence of an annulus. Microscopically, thin sections were prepared and examined at 1000x oil immersion, revealing broad ellipsoid basidiospores, clavate basidia, and clamp connections. Molecular identification targeting the rDNA-ITS1/ITS2 regions revealed that the Kendari specimen (ITSPV719613) clustered with A. javanica from Indonesia and formed a sister clade to A. javanica from Malaysia, consistent with the BLAST result showing 100% and 94,59% similarity, respectively.
